Unusual because it is encoded in mitochondrial DNA rather than in the cell nucleus. Research treats it as a signal that mitochondria send to the rest of the cell about metabolic state.

MOTS-c is a sixteen amino acid peptide encoded within the mitochondrial genome. That origin is the reason for the research interest: it is evidence that mitochondria participate in signalling rather than only in energy production.

Where it came from

Identified in 2015 by Lee and colleagues at the University of Southern California, working on short open reading frames within mitochondrial DNA. It is one of a small set of mitochondrial derived peptides described since.

How it is thought to work

Proposed to act on the folate and purine biosynthesis pathway, with downstream activation of AMPK, a central sensor of cellular energy state.

Some work reports MOTS-c translocating to the nucleus under metabolic stress and influencing transcription there, which would make it a genuine retrograde signal from mitochondria to nucleus.
